Amoeba may offer key clue to photosynthetic evolution

ScienceDaily (Feb. 27, 2012) ? The major difference between plant and animal cells is the photosynthetic process, which converts light energy into chemical energy. When light isn't available, energy is generated by breaking down carbohydrates and sugars, just as it is in animal and some bacterial cells. Two cellular organelles are responsible for these two processes: the chloroplasts for photosynthesis and the mitochondria for sugar breakdown. New research from Carnegie's Eva Nowack and Arthur Grossman has opened a window into the early stages of chloroplast evolution.

Their work is published online by the Proceedings of the National Academy of Sciences in the week of February 27-March 2.

It is widely accepted that chloroplasts originated from photosynthetic, single-celled bacteria called cyanobacteria, which were engulfed by a more complex, non-photosynthetic cell more than 1.5 billion years ago. While the relationship between the two organisms was originally symbiotic, over evolutionary time the cyanobacterium transferred most of its genetic information to the nucleus of the host organism, transforming the original cyanobacterium into a chloroplast that is no longer able to survive without its host. A similar process resulted in the creation of mitochondria.

To sustain the function of the organelle, proteins encoded by the transferred genes are synthesized in the cytoplasm, or cell's interior, and then imported back into the organelle. In most systems that have been studied, the transport of proteins into the chloroplast occurs through a multi-protein import complex that enables the proteins to pass through the envelope membranes that surround the chloroplast.

Clearly the events that gave rise to chloroplasts and mitochondria changed the world forever. But it is difficult to research the process by which this happened because it took place so long ago. One strategy used to elucidate the way in which this process evolved has relied on identifying organisms for which the events that resulted in the conversion of a bacterium into a host-dependent organelle occurred more recently.

Nowack and Grossman focused their research on a type of amoeba called Paulinella chromatophora, which contains two photosynthetic compartments that also originated from an endosymbiotic cyanobacterium, but that represent an earlier stage in the formation of a fully evolved organelle.

These compartments, called chromatophores, transferred more than 30 of the original cyanobacterial genes to the nucleus of the host organism. While gene transfer has been observed for other bacterial endosymbionts, the function of the transferred genes has been unclear, since it does not appear that the endosymbionts (in contrast to organelles) are equipped to recapture those proteins, because they do not have appropriate protein import machineries.

The Carnegie team honed in on three of the P. chromatophora transferred genes, which encode proteins involved in photosynthesis, a process localized to the chromatophore. They set out to determine whether these proteins are synthesized in the cytoplasm of the amoeba and whether the mature proteins became localized to the chromatophore.

Using an advanced array of research techniques, they were able to determine that these three proteins are synthesized in the cytoplasm and then transported into chromatophores, where they assemble together with other, internally encoded proteins into working protein complexes that are part of the photosynthetic process.

Interestingly, the process by which these proteins are transported into chromatophores may also be novel and involve transit through an organelle called the Golgi apparatus, prior to becoming localized to the chromatophore. This suggests the occurrence of an initial, rudimentary process for proteins to cross the envelope membrane of the nascent chloroplast. This process ultimately evolved into one that is potentially more sophisticated and that uses specific protein complexes for efficient transport.

"This work demonstrates that P. chromatophora is a potentially powerful model for studying evolutionary processes by which organelles developed," Nowack said. "Obtaining a comprehensive list of proteins imported into chromatophores, including their functions and origins, as well as understanding the pathway by which these proteins are imported, could provide insight into the mechanism that eukaryotic cells use to 'enslave' bacteria and turn them into organelles such as chloroplasts and mitochondria."

Wakefield Rhubarb, food and drink festival

This weekend is the annual rhubarb, food and drink festival held in Wakefield.It's a great event to attend, lots to see and do, plenty of free foods to sample and lots of local food producers selling their wares.Oldroyds farm in Carlton,Lofthouse, is the UK's leading producer of Yorkshire forced rhubarb and you can actually go on tours of the rhubarb sheds and it is reputed that you may even be able to hear the rhubarb growing! I haven't been on the tour so I can't vouch for that but I do like rhubarb, being a fairly recent convert, so we decided to pop along and see what was on offer.
You can just make out the lovely cathedral spire behind the food demonstration marquee.


As you can see we bought a lot of rhubarb products, not all are for us though, we got some things for our parents too.I find cupcakes to be quite sickly (I know they are all in vogue but I find them a bit claggy sometimes, all that gooey icing!) but the rhubarb in the cake mix was just sharp enough to cut through the sweetness and they tasted divine! We couldn't resist sampling the local beer, I am a real ale fan anyway and thought the rhubarb bitter was just gorgeous.You could smell the fruit in the beer and it had a real kick to it.
Even the pork pie was delicious (and I do not profess to be a big pork pie fan) with the hint of rhubarb.

The company's current top of the range handset is the Viewsonic V350 which is also a dual SIM phone with a five megapixel camera and Android 2.2 Froyo and retails for around ?170.

To investigate the possible roles of Bcl-2 family during the colorectal cancer development as well as to find the new targets for further therapy, we detected the expression of Bcl-2, Bcl-XL and Bax in tumor tissues and adjacent tissues for patients with colorectal cancer at both mRNA and protein levels. Our data demonstrated that Bcl-XL was dramatically higher in tumor tissues than in adjacent tissues at both mRNA and protein levels, whereas other two genes had similar expression between tumor and adjacent tissues. Next, results also showed that Bcl-2 protein was negative correlation with lymphatic metastasis and Dukes stages, it was a risk factor for overall survival as well as tumor-free survival, lack of Bcl-2 expression showed a worse prognosis. However,there is no relationship between the expression of Bcl-XL and Bax protein and clinicopathological parameters including prognosis. Furthermore, we treated DLDl cells with an artificial synthesized double-strand small interfering RNA targeting Bcl-XL (Bcl-XL siRNA). Results indicated that the level of Bcl-XL protein in DLDl cells was down-regulated by Bcl-XL siRNA effectively, and the apoptotic ratio induced by Bcl-XL siRNA was dramatically higher than that caused by control treatments , whereas the survival rate was decreased significantly.To sum up, the following conclusions could be drawn out from this study:1) Bcl-XL expression at both mRNA and protein levels were much higher than Bcl-2, suggesting it might be more important for colorectal cancer and it might be a good therapeutic target.2) Bcl-2 protein might be an indicator for prognosis evaluation.3) Bcl-XL siRNA could effectively induce the apoptosis in human colon cancer cells, suggesting it might be a new strategy for colon cancer.

Under the cover of the law, we live and act as our forefathers did under the rules of divinity or the protection of the monarch.

The report recommends that federal, state and territory governments, in consultation with the non-government school sector, should make reducing educational disadvantage a high priority in a new funding model, with resourcing to be targeted towards the most disadvantaged students.

Gonski and co identify ?five factors of disadvantage that have a significant impact on educational outcomes in Australia?. They say: ?At the student level these factors are socioeconomic status, Indigeneity, English language proficiency, and disability. At the school level, remoteness is demonstrated to have an impact on student outcomes.?

The inequities that the report identifies in education are related in many ways to the inequities that occur in the distribution of health, as well as in access to healthcare.

Its recommendations aim to ensure that ?differences in educational outcomes are not the result of differences in wealth, income, power or possessions? and that ?all students have access to a high standard of education regardless of their background or circumstances.?

From a perspective of wellbeing we might look for progress in areas such as young people:

? ???????understanding their social, emotional and physical being in ways which enable them to look after themselves and their friends
? ???????having a sense of the evolving history and culture of the country in which they live

? ???????being well placed to take part in the working, social and cultural life of their communities.

From the school system we might look for progress towards:

From a health perspective, the Gonski review should provide some focus on the social determinants of health and on preparing young people with life skills and an appreciation of lifelong learning.

While the focus appears to be on measuring literacy in maths, science and English, we are now seeing generations that have had little background in supportive stable home environments on which they might model their future behaviours.

The value of relationships, healthy activity, fitness, lifestyle, good nutrition and productive effort need to be seen as equally important.

There are many good examples of these endeavours already in place in our schools but a funding system that focuses purely on performance measures on literacy needs to not only be avoided, but should be an opportunity to highlight the importance of these fundamental educational objectives in building good lives and opportunities for young people.

In the end they will be our future leadership and their education and socialisation will determine what sort of a health system we need.

Too much concentration on ideology around public/private funding will obscure the importance of these more fundamental issues.

Young people?s health and wellbeing does not only depend on how they personally respond to social, economic, cultural and environmental issues in terms of their own choices and behaviour and how these shape their health, but also on their role as citizens or social agents in changing those conditions to improve population health.

This broader, social role has particular relevance to the cross-curriculum priorities (sustainability, Asian studies, Indigenous studies) and how they might bear on, and incorporate, young people?s health and wellbeing.

Indeed, students? health and wellbeing could provide an integrating framework for the whole curriculum.

Population health:

?.Acknowledging the importance of culture and mental health highlights the social significance of health in two ways: by casting new light on human development and national progress, and by showing health is an important social dynamic: a cause as well as a consequence of social change and people?s resilience. A broader view of the science and politics of population health would not only benefit health directly, it would allow public health to play a more influential role in public and political debate about national and global issues and priorities, including sustainable development, so having a more indirect, but ultimately powerful, effect on people?s health and wellbeing.

Sustainability:

?. Population health perspectives can make an important contribution to sustainable development: they provide a means of integrating, balancing and reconciling different social priorities by allowing them to be measured against a common goal or benchmark: improving health and wellbeing. Population health is, then, a key element of the quest for a socially, economically and environmentally sustainable way of living ? humanity?s greatest present challenge.

?.A broad view of population health and its social determinants ? socio-economic, cultural and environmental ? challenges the legitimacy of the dominant worldview or paradigm of material progress (which gives priority to economic growth and a rising standard of living), and supports the alternative, sustainable development (which seeks to balance and integrate social, economic and environmental goals to achieve a high, equitable and enduring quality of life). The contest between the two models, or narratives, of progress has been framed in economic and environmental terms, and the social dimension has been neglected. Population-health research can help to correct this distortion.

For example, it is clear that material progress does not simply and straightforwardly make people richer, so giving them the freedom to live as they wish. Rather, it comes with an array of cultural and moral prerequisites and consequences (for example, giving priority to money and what it buys) that affects profoundly how people think of the world and themselves, and so the choices they make. The pursuit of material progress, of evermore enrichment, as a cultural (and political) priority is jeopardising global health in ways that go well beyond issues of socio-economic disadvantage and inequality.

Thus the core social challenge is not primarily a poverty of the means to the end of ?the good life? as it is currently defined and pursued; it is a poverty of the end itself. Giving the poor and marginalised the opportunities and privileges of the majority, however much it will help them, will not solve the problems of population health; nor will reducing inequality. This is especially true in the developed world, but it also applies to developing countries such as China and India, which have a rapidly growing middle class that is increasingly susceptible to diseases of affluence and modernity. Worldwide there are over 1.3 billion people who are overweight and about 800 million who are underweight, and the numbers are diverging rapidly.

Indigenous studies:

?. Sveiby and Skuthorpe suggest in Treading Lightly that traditional Aboriginal society provides a model or recipe for sustainability. But the word that came to my mind in reading the book was that this was a parable or allegory for our times, an extremely important one. The moral is not that we could or should adopt an indigenous lifestyle, but that we need to recognise that other, quite different, and even better, ways of making sense of the world and our lives are possible. And not only that: we need to examine our present situation at this most fundamental level if we are to have any chance of achieving a high, equitable and durable quality of life.

Asian studies:

?other evidence for the need to look beyond happiness scores comes from studies that suggest collectivists have a higher tolerance for contradictions, cope better with change, are more flexible across social situations, and think more holistically. East Asians exhibit ?holistic? cognition, characterized by paying attention to the entire stimulus field and the use of dialectical reasoning, while Americans exhibit ?analytical? cognition, characterized by paying attention to isolated details and the use of Aristotelian-type logic. East Asians focus more on relationships and connections, on fitting in rather than standing out. Being less concerned with self-esteem, they are more willing to appraise their strengths and weaknesses, to address their shortcomings and to persevere in the face of difficulties. These differences have obvious implications for how societies deal with modern conditions and challenges. Even the most cursory assessment of China and the United States today exposes the danger of using happiness as a measure of their national performance.

Research commissioned by the panel found that one-third of all Australian schools serve a student population where the average socioeconomic background of students is below the national average.

Enrolment data also confirms a steady drift of students from the government sector to the non-government sector that has been evident since the late 1970s, as well as a tendency for parents from higher socioeconomic backgrounds to move their children from government schools to non-government schools or select-entry government schools.

Almost 60 per cent of the decline in government school enrolments between 1975 and 2006 occurred in the top half of the socioeconomic status distribution.

Government expenditure on schooling in Australia is relatively low in comparison to other OECD countries. The OECD estimates that public expenditure on primary, secondary and post-secondary non-tertiary education in Australia was 3 per cent of gross domestic product (GDP) in 2008, compared to an OECD average of 3.5 per cent of GDP.

In terms of private expenditure on primary, secondary and post-secondary non-tertiary education, Australia is above the OECD average, with private expenditure equating to 0.6 per cent of GDP in 2008, compared with the OECD average of 0.3 per cent of GDP. These data can be explained in part by Australia?s large non-government school sector.

The number of Australian students who attain a Year 12 or an equivalent qualification has progressively increased over the past decade. Sony has finally come out with its replacement for the PlayStation Portable, and it's impressive. The PlayStation Vita takes the PSP's signature design and adds more processing power, optional 3G, dual analog sticks, and an amazingly bright, colorful OLED screen. The result: A handheld gaming device that eclipses the PlayStation 2 in power, and comes very close to the capabilities of the PlayStation 3. Sony made some strange decisions with the Vita's media playback abilities and Web browser, but for gaming it's incredible, making it our Editors' Choice for handheld game systems, and a significant step ahead of the Nintendo 3DS (4 stars, $169.99).

Design
Out of the box, the PS Vita looks like the PlayStation Portable at first glance, but the overall build quality of the PS Vita is much better, right down to the small details. Without a pop-out door for UMDs or a sliding design, the PS Vita feels much more solid than the PSP, and despite their small size the analog sticks have just enough give for responsive control without feeling loose. Besides the proprietary USB port and headphone jack on the bottom, every port and slot on the PS Vita is covered by a door. The top edge holds the game card slot and an accessory port, the bottom edge holds the memory card slot, and the left edge holds the SIM card slot. The handheld weighs a solid 9.8 ounces, and at 7.2 inches wide by 3.3 inches tall by 0.7 inches deep it's almost the same as the PSP in shape.

Sony PlayStation Vita

The layout is pure PlayStation, with a direction pad, four face buttons, Start and Select buttons, a PlayStation button, two shoulder buttons, and two analog sticks. The top edge of the PS Vita holds the power button and volume controls. While there are only two shoulder buttons instead of four like on the PlayStation 3 controller, the front touch screen and back touchpad have enough input for no-compromise control, unlike the PSP with its single analog pad. The back touchpad sits seamlessly on the back of the PS Vita, without a frame or bezel, while two rubberized grips give your fingers a comfortable place to rest when you're not using the trackpad.

Hardware
The 5-inch OLED touch screen is gorgeous. It's bright, colorful, and extremely sharp. Its 960-by-544-pixel resolution makes it almost exactly equivalent to many current-generation large-screen Android phones, and not quite as high-resolution as the iPhone 4S or several 720p Android phones on the market. But at five inches, the PS Vita's display is more than sharp enough to lend a satisfying impression of high-definition video to a movie or game. The screen is also multitouch, so you can pinch to zoom in the Web browser, the map software, and in games.

The front- and back-facing cameras seem identical, and are only slightly sharper than the cameras on the 3DS. The Vita might compare favorably to the camera phones of a few years ago in image quality, but if you have a decent smartphone, there is no reason to use the Vita's camera for anything besides video chat and AR (augmented reality) games.

As a nice touch, the PS Vita supports Bluetooth devices just like the PS3, including stereo headsets. I paired my Logitech Wireless Headset to the Vita and could hear the background music of the menus and the dialog in Uncharted: Golden Abyss easily. If you don't have a Bluetooth headset, the PS Vita still has a 3.5mm headphone jack and on-board stereo speakers.

Spanish football player dies from heart failure

MADRID (AP) -Spanish news agency Efe has reported that a 21-year-old Spanish football player has died from heart failure suffered after a training session.

Luis Miguel Lastra was found late Friday in the stairwell of his home following practice with his club Ciudad Jardin, of the Andalucia first division in a regional league.

Ciudad Jardin president Jose Bonilla told Efe the player's death was "natural, as a result of heart failure."

Lastra's death follows deaths of other Spanish football players from heart-related problems in recent years, including Espanyol captain Daniel Jarque and Sevilla's Antonio Puerta.
