Yemeni government official escapes suicide attack

SANAA, Yemen (AP) ? Yemeni security officials say a government official who once led a local militia against al-Qaida in the country's south has survived a suicide attack. One of his guards died of wounds sustained in the attack.

The officials said the attacker detonated his explosives Saturday near the vehicle of Mohammed Aidarous, the head of Lawder district in Abyan province. The officials said the explosion left 10 wounded, including three civilians. One guard later died of his wounds in a hospital in Aden. Aidarous had led a local militia which fought alongside Yemen's military to drive al-Qaida militants out of their district.

It was the fourth assassination attempt against Aidarous, who was unharmed.

The officials spoke on condition of anonymity because they were not authorized to speak to the media.

Applying These Tips Could Help You Repairing Your Credit. | Bob ...

Is your credit terrible? The helpful advice in this article will help you find a plan to transform your credit report into one you will be proud of.

Make sure to keep a record of your communication with credit bureaus in case they make mistakes. Keep a record of all your phone calls and correspondence. Have your dispute letter certified so you have proof of it both being mailed and being received.

Paying off each of your accounts and bills on time can help to repair your credit. Credit counseling services could help.

TIP! When you are trying to repair your credit, be realistic. Some credit reporting agencies may try to get you to commit to an unrealistic payment schedule, which will throw you further in the hole.

When you start fixing your credit, pay down balances on the credit cards as soon as you can. Pay off high-interest debt first, as it grows the fastest. Beginning to pay your credit card balances off will show creditors that you are making a valiant effort and are credit worthy.

Debt Consolidation

Consider debt consolidation as a possible tool to help you repay your debt and rebuild your credit record faster. Consolidating your debts may be the best way for you to reduce your debt and fix your credit. This way all of your debts will be combined into one bill and it will be easier to pay. When considering debt consolidation you will need to ensure that you can afford the payment amount.

Building your credit back up is the number one way to fix a bad credit score. Prepaid credit cards are great for this, since you can get improved with a bad credit score, and there?s no way to rack up debt and late fees. A potential lender will see this as a responsible move on your part, showing that you are credit worthy.

TIP! Opening additional lines of credit will negatively affect your credit score. Avoid obtaining checkout credit cards that offer huge discounts.

If you are having a difficult time creating or sticking to a budget, get in touch with a responsible credit counseling organization. Often times, these companies will work with the creditors to get negotiate a lower interest rate and an affordable payment plan that will help get your finances back on track. Credit counseling can be a key piece in helping you understand how to budget your salary and pay your bills.

Do not use your credit cards. Pay with cash whenever possible. Any credit card purchases should be paid in full the same month of purchase. Do not carry a balance on your cards.

Credit Score

Financing a home can be difficult if you have bad credit. If you do have poor credit, try to get a FHA loan because there is a guarantee that it will be given to you. Even when the resources for making down payments or paying closing costs are lacking, FHA loans can help.

TIP! Pay all outstanding debt to begin repairing your credit. Your credit will only get worse if you are not paying off your debts.

In order to fix your credit, you need to have a plan to pay debts off. Your credit score is damaged by large amounts of existing debt along with a history of late payments. Budget realistically, and set aside as much as possible to pay towards your outstanding debt. Having no current debt will help to raise your credit score.

Instead of avoiding collectors, talk to them to see what kind of payment arrangements they can offer you in light of your financial situation. Tell the debt collector the amount that you can pay them, and when they can expect the payment. Collection agencies are usually willing to negotiate.

Read your negative reports carefully when attempting to rebuild your credit. There may very likely be errors or mistakes that can be removed.

No credit repair company can remove factual information, no matter how damaging, from your credit report. You have to wait for seven years before negative data can come off your record. You should know that mistakes and anything incorrect can be removed from your credit report.

You should know of any inquiries about your credit score. Anytime someone looks at your credit score, that inquiry is noted.

If you have to improve your credit, make a solid plan and follow it. You must make a commitment to making changes on how you spend money. Sticking to necessities for a while is crucial. Ask yourself how necessary each purchase is, and how affordable it is too. If you can?t answer ?yes? to each of the questions above, you need to reconsider the purchase.

Credit Report

Go through your credit report with a fine tooth comb before resolving to accept the issues as valid. Credit reports sometimes contain inaccurate information. A credit dispute can be initiated in these cases; they take time to be resolved, but the discrepancy is rectified if found to be in error.

TIP! This is one of the quickest way to create and maintain a credible financial record. Late payments to credit cards are reported to the major credit agencies and can hurt your chances for securing a new loan.

As you?ve now learned, a credit report doesn?t have to be the stuff of nightmares. You don?t have to be afraid of your credit score; you can improve it. Just follow these tips we listed, and you will have the credit report that will be the envy of all of your friends

Welcoming Your New Life With Unlimited Potential | Rob Moore ...

Personal development requires that you educate yourself. This will ensure that your personal growth will be efficient. There are tons of ways to jumpstart your personal growth, and below you?ll find some excellent ones with which to begin.

Make a decision about where you want to go in life and stick with it. Sitting and thinking about the life you want will not get you that life. Identify your goals and take the necessary actions to make it happen.

Recognizing your limits when performing physical tasks can actually help you meet your goals. While exerting your best effort is admirable, recognize when you have reached your limit. You need to be careful about your physical health even while you are striving to achieve your goals. If you sacrifice the needs of your body, it will greatly hinder your self improvement.

To become a champion, you need an excellent coach. Someone to help guide you is always helpful. Everyone who has ever been good at anything has had help from a coach or mentor. Every person should help others find the way. Without someone to teach them the right way of doing things, it?s impossible to expect them to be able to properly succeed. Everyone who is a great success had a great coach.

You can?t please all of the people, and there?s no use in trying. Please yourself first! This does not mean privileging your happiness to a fault, but it does mean that you must take responsibility for your own happiness. If what you?re doing is true to both your ethics and yourself, then you can be healthier with your path to personal development.

When you are pursuing your personal development goals, having setbacks can be discouraging and make you feel like a failure. Failure is just a learning experience incognito. It shows you what your strengths and your weaknesses are. So, in a way, each failure can be viewed as an opportunity to gather important information of use in your personal development efforts.

If you have extensive personal development goals, consider enlisting the help of a therapist. While self-help books help to a certain degree, seeing a therapist has more personalized substance to it. Sometimes, the best solution comes about through the simple act of talking with a trained therapist. Books can?t talk back like a trained professional, after all.

Encouraging faith by love will lead to a higher level of self improvement. Faith cannot exist without love. You cannot consider your faith satisfactory if you do nothing to express it. Help out those in need, and make sure that your faith really means something by showing love to your peers and yourself.

Perhaps stress is keeping you from feeling happy. You are harming yourself mentally and physically when there is too much stress in your life. Eliminating stress is essential for thinking clearly and achieving goals. Make time in your schedule each day to take a few minutes alone and clear your mind. Having a time to refresh can give you peace and improve your self-image.

Moving toward your goal and meeting your personal needs demands that you do one specific thing to be successful. It is important to play an active role in your own life story, instead of just watching it pass by. Sitting back and watching will make you dull and unhappy.

Dealing with situations rationally is a skill that you can learn from others or teach yourself. Stay graceful under fire, and you will gain confidence that helps in your day to day life. Take the time to reflect on the situation, breathe deeply and relax.

If you are anxious about social interactions, try just going to a movie with a friend. You will be out and about but you don?t have to talk a lot or even see who you?re with most of the time. Eventually, it will be second nature to be around a large amount of people.

Every morning when you wake up, tell yourself that you are going to make today better than yesterday. Try to see each day as a way to improve. Set a goal to do something you couldn?t do yesterday, or improve on something you were able to do.

Getting Rid

By being a leader, you can help improve your personal development. Leadership has many definitions, but many people think of it as ?influence.? Take a look at your leadership journey. Are there events that have had major impacts on your life? How did the events that took place have an effect on you? Which one of your attributes is most influential in making you a good team player? If you explore the possible answers to these inquiries, you will become more capable of functioning effectively in a team setting.

Are you an alcohol abuser? Are you a smoker, or do you regularly engage in something that is bad for you? You should treat your body like a temple, because that?s what it is. Try getting rid of your bad habits; it can be important to making your life better. Look at your life, and decide if you have any bad habits that you need to work on getting rid of.

Treat everyone with respect, no matter what they can do or how much power those people may have. The way you behave toward others has more to do with your personality than with anything they say or do.

Instead of talking up all of your trophies and accomplishments, ask other people about their notable achievements and proudest moments. You?ll be able to find out about the wonderful things the people you know have accomplished, bringing about a new-found respect and admiration for them.

You should know the values that bear importance to you prior to planning a personal development program. Focus on those areas first to make the positive changes that are most important to your happiness. Instead, work on aspects of your life you want to improve that mesh well with your individual beliefs. This will help you make significant changes to your life, both at home and at work.

Having a better life often happens because of willpower, and keep in mind that the things you wish for others might have an effect on you. If you seek a healthy, happy environment, make sure to provide one for others. If you try to remain positive in all aspects of your life, you will not be as likely to get dragged down by sadness, anger and other negative feelings.

One part of depression you may not thought of to look at is your diet and increasing the amount of complex carbohydrates that you consume. A lack of complex carbs can lead to a shortage of serotonin, a significant factor in depression. You can do this by eating more raw veggies, fruits, nuts, brown rice, whole grains and assorted beans.

You should create a list of goals as part of your self improvement efforts. One thing a lot of people lack and what you might want to write down is to have more confidence. The next step is to seek different ways to grow that skill or trait. Once you know what you need to do, you need to put yourself in an environment where you make those things happen. When you tackle it methodically, you give yourself a better chance at succeeding.

You should try to find and read books that tell you about personal development. The best personal development books can give you new ideas and advice that could change your life forever. Try to pick a volume that has already been well-received by others because poorly done books in this category are fairly common.

As stated in the introduction, personal development consists of expanding your knowledge. Gaining this knowledge is important, but it must also be applied. The avenues open to you for self-improvement are many. Your confidence should improve if you apply some of the tips mentioned in this article

Find texts that help you. Some people may look to a religious text, while others might look for inspiring quotes. You may need help staying on top of life and keeping your mind clear, and a trusted book whose words have meaning to you is a great resource to fall back on.

Refs cheered at first NFL game following lockout

BALTIMORE (AP) ? No one is complaining that the refs cost the Cleveland Browns the game. That mere fact is a major victory for the NFL and the seven-man crew led by referee Gene Steratore, who brought official harmony back to the nation's most popular league.

Cheered from the moment they walked onto the field, the men in stripes ran a smooth and efficient game Thursday night as the NFL's lockout of officials came to an end with the Baltimore Ravens' 23-16 win over the Browns.

"To just be applauded by 50,000 people prior to anything happening, it was something that kind of chokes you up," Steratore said. "It was a very special feeling."

Sure, there were calls that made both sides unhappy. Browns coach Pat Shurmur drew an unsportsmanlike conduct penalty for arguing an intentional grounding call, and Ravens left tackle Michael Oher could be heard raising all kinds of beef about a holding call.

But, overall, the officials kept the game in control, curtailing the chippy play and choppy pace ? not to mention the inconsistent calls ? that had marred the three weeks of games with replacement officials.

"It was great to have those guys back," Ravens running back Ray Rice said. "It looked like they knew what they were doing."

An agreement to end the lockout was reached late Wednesday after marathon negotiations, two days after a "Monday Night Football" finish brought debate over the use of the replacements to a fevered pitch nationwide.

That game ended when a 24-yard desperation pass on the last play was ruled a touchdown ? even though replays appeared to show it should have been an interception ? giving the Seattle Seahawks a disputed 14-12 victory over the Green Bay Packers.

The stage was set for something eerily similar Thursday. A fourth-down unnecessary roughness penalty on Baltimore's Paul Kruger ? a good call, given the way he shoved Cleveland's Joe Thomas after the whistle ? gave the Browns one final play from the 18-yard line.

But Brandon Weeden's 18-yard pass sailed high as time expired. No controversial ending this time.

"I thought they handled (the game) great," Cleveland coach Pat Shurmur said. "I had all the confidence in the world that this was going to be officiated in the right way."

The newfound love for the refs was evident all evening.

About an hour before kickoff, they made their first appearance on the field and heard cheers from the early arrivals. A few minutes later, Steratore was shaking hands with Shurmur near midfield and getting a hug from Ravens face-of-the-franchise Ray Lewis at the 30-yard line.

Later, when the crew returned, they received a standing ovation and doffed their caps to the crowd. One fan held up a sign that read: "Finally! We get to yell at real refs! Welcome back!"

"It was very chaotic with the replacement refs," said Karen Riley, a 44-year-old fan wearing a Rice jersey. "They couldn't control the players on either side. There were bad calls, constantly, and in some cases refs making different calls at the same time."

When Steratore then turned on his microphone to greet the captains for the pre-game coin toss, the crowd heard him say: "Good evening, men. It's good to be back."

The stadium erupted in a roar.

"You know we always pride ourselves in being a face without a name," Steratore, a 10-year league veteran, told The Associated Press about an hour before kickoff. "This will be a little different, but I don't expect it to last too long. And that's the goal ? is that we can let them get through that portion of this. It's happy to be back, it's happy to be appreciated. But then as soon as the game starts, it's happy to disappear again and let the entertainers entertain."

The deal to end the lockout is only tentative ? it must be ratified by 51 percent of the union's 121 members in a vote scheduled for Friday and Saturday in Dallas ? but both sides nevertheless went forward with the plan to have the regulars back for Thursday's game.

So Steratore hustled to Baltimore, making the 3?-hour drive Thursday morning from his home in the Pittsburgh area. He's usually in place the day before a game, but none of his regular pregame meetings had to be changed because the Browns-Ravens game was at night.

"We've had a few weeks to actually realize that this was the first September that I was home for multiple Saturdays and Sundays for almost 30 years of my life, continuously. ... It just feels completely different," Steratore said. "To be away from something that is involved with this level of professional sport, just to come back and feel that again, it doesn't take long to realize why you were missing it as much as you were missing it."

Steratore, who is a basketball official in the Big East Conference among others, also was fully aware he would be jeered the first time his crew made a questionable call ? just like always.

"Without a question," he said. "I've been yelled at by my own children many times, so this won't be any different."

Sure enough, the same fans that cheered the coin toss let out a full chorus of boos when line judge Jeff Seeman tossed his yellow flag some 20 yards to whistle Baltimore safety Bernard Pollard for a personal foul in the third quarter. Replays showed it was a good call: Pollard led with his helmet to make contact with a defenseless receiver, costing the Ravens 15 yards in a drive that led to a field goal for the Browns.

Steratore's crew nearly made a misstep in the first quarter, incorrectly spotting the ball by 2 yards after a misapplication of the rules following a holding call on the Browns. But two members of the crew caught the mistake and notified the referee before the next snap. A brief huddle ensued, and the ball was moved to its correct spot.

The crew made it clear it wouldn't tolerate the extra shoving and yelling after the whistle that had been frequently permitted by the replacements. Offsetting personal fouls were called on Cleveland's Johnson Bademosi and Baltimore's James Ihedigbo for extracurricular roughness on a punt return in the first quarter.

Then there was Shurmur's unsportsmanlike conduct penalty. Replays appeared to validate the grounding call he was trying to contest, and the coach took responsibility for his loss of temper.

"I can't do that," Shurmur said. "It's an emotional game, and I got to make sure I keep my emotions in check."

There were 18 penalties called in the game, mostly the familiar calls for holding and false start. There were two rare ? and indisputable ? whistles for fair catch interference on punt returns, and a hands-to-the-face call on Baltimore's Kelechi Osemele was so obvious that it drew three flags.

The league's new agreement with the officials runs for eight years. Commissioner Roger Goodell acknowledged the ending of the Seahawks-Packers game "may have pushed the parties further along" in the talks.

"Obviously when you go through something like this it is painful for everybody," Goodell said. "Most importantly, it is painful for our fans. We are sorry to have to put our fans through that, but it is something that in the short term you sometimes have to do to make sure you get the right kind of deal for the long term and make sure you continue to grow the game."

The dispute even made its way to the campaign trail, with President Barack Obama's spokesman, Jay Carney, calling Thursday "a great day for America."

"The president's very pleased that the two sides have come together," Carney said.

Microsoft Sculpt Comfort Keyboard

The Microsoft Sculpt Comfort Keyboard ($59.99 list) retains the same distinctive curvy design featured in some of Microsoft's earlier contour keyboards, and comes with some new features like a detachable padded wrist rest and Windows 8-specific charm keys. The most striking change that's surely going to attract the most attention, though, is its dual-function spacebar, which is an otherwise ordinary spacebar (albeit considerably wider) that has been split into two separate keys, with the right half acting as a dedicated spacebar while the left side can be toggled between serving as a spacebar or a backspace key. Apart from these elements, though, there's not much to distinguish the Sculpt from other keyboards currently on the market.

Design and Features
The Sculpt is full-size keyboard with an all-black chassis constructed entirely out of plastic. The only departure from the Sculpt's plastic build is its detachable padded wrist rest, which can be removed by unlocking a latch on the underside of the keyboard. With the exception of a thin glossy strip running along the outer perimeter of the frame, the Sculpt sports a matte-finished look. Aside from giving the keyboard subdued, elegant look, the matte finish does a good job of resisting smudging after prolonged use, unlike the glossy-framed Logitech Wireless Solar Keyboard K750 . The Sculpt doesn't sport a thin profile like the K750, as its frame has a gradual bulge that raises its profile around the crescent of the wavy contour. The rounded design continues on Sculpt's scalloped keys, which feature a slight curvature on the bottom.

A switch on the upper right hand of the Sculpt allows users to toggle the top row of keys between standard function keys and a mixture of Windows 8-optimized hotkeys and charm keys. I used the Sculpt on a system with Windows 8 Release Preview installed and was able to dig a little deeper into its interface. Accordingly, the Sculpt's F1 through F4 keys are standard media playback keys that, incidentally, are compatible with earlier versions of Windows. Meanwhile, the F5 through F8 keys are shortcuts to Windows 8's five so-called "charms," which are share, settings, search, a device toggle, and a desktop button; the desktop button is also a charm, but that particular key, which features the new Windows logo, is on the Sculpt's lower left side. Without a Sculpt, you can accesses the charms by swiping from the right edge of a touch-enabled display.

The F9 through F12 keys, meanwhile, act as Windows 8 hotkeys that pertain to apps; F9 rotates between open apps, F10 lists all open apps on the left side of the screen, F11 enables a split-screen mode between any two apps, and F12 lists all open apps on the bottom portion of the screen. While these Windows 8-optimized keys succeed in making Windows 8 a more mangeable interface, the Sculpt's directional arrows unfortunately don't scroll through the tiled interface of the Windows 8 desktop screen, and instead simply allows for moving between individual tiles. Ultimately, this means that even with a Sculpt keyboard and a non-touch-capable system, you'll still need to use a compatible mouse to fluidly navigating the tiles?the Sculpt alone, alas, won't get you there.

And now, the spacebar. Microsoft's stated rationale for its decision to split the spacebar in half is to eliminate so-called "pinky reach," or the purportedly unnatural movement that occurs when one's pinky strikes the backspace key. It's premised on the notion that one can type more efficiently while also reducing strain by backspacing with the left hand's thumb. Fortunately, Sculpt users have the option to toggle between the dual-functionality approach or a traditional setup wherein both halves act as a spacebar by simply holding both keys for several moments.

The Sculpt uses two AAA batteries (which are included). If you're the type who constantly worries about battery life, then you'd likely be better off with either the solar-powered K750 or the Logitech Wireless Illuminated Keyboard K800 , which can be recharged via USB connection. The Sculpt is compatible with Windows 8, Windows RT, Windows 7 and Vista, though the Windows 8 charm keys and most of the hotkeys (with the exception of the media playback keys) don't all work with earlier versions of the operation system, save for the volume control and application selector.

Performance
Unlike the Microsoft Bluetooth Mobile Keyboard 6000 , the Sculpt connects wirelessly via an included USB dongle. If you plan on taking it on the go, be sure to remember where you place the dongle, as the Sculpt doesn't have any compartments on its underside for storing the dongle, unlike the Logitech Logitech Wireless Keyboard K360 . Unlike Logitech's proprietary "unifying" receiver technology, which allows up to six of its peripherals to connect via the same dongle, the Sculpt requires an entire USB port to itself, which may be problematic for users with systems with a limited amount of USB ports, like ultrabooks.

Typing on the Sculpt makes for an overall comfortable experience, especially when the wrist rest is attached. Moreover, the contoured shape curves in accordance with the natural posture of your hands. The response rate was on point; there was no lag time whatsoever. Throughout testing, the pitch of the keys felt just right, and they felt agreeably springy whenever bottoming out at the end of a keystroke.

While I've never considered "pinky reach" a particularly pernicious threat, I nonetheless was eager to try out the spacebar's dual-functionality feature. Overall, I had a difficult time adjusting to it, and found that it decreased my productivity. Of course, this is a very subjective call that ultimately boils down to taste, so you'd be better off trying it for yourself. Personally, I liked the Sculpt much more when the dual-functionality was turned off.

The Microsoft Sculpt Comfort Keyboard is a solid choice for anyone who wants a keyboard optimized for Windows 8 or who can envision a smoother typing experience when there's a backspace button next to the spacebar. Aside from these considerations, though, it's a fairly standard keyboard that lacks some of the more common features seen in other keyboards, like backlighting or a rechargeable battery. For these reasons, it doesn't have enough to dethrone our current Editors' Choice for keyboards, the Logitech Wireless Illuminated Keyboard K800.

Bayer wins early U.S. approval for colon cancer drug

(Reuters) - The U.S. Food and Drug Administration on Thursday approved a new drug for advanced colon cancer developed by Bayer AG, a month ahead of the agency's expected action date for a decision.

The drug, to be sold under the brand name Stivarga, was approved to treat colon cancer that has progressed after prior treatment or that has spread to other parts of the body, the agency said.

"Someone has clearly lit a fire under the FDA," Sanford Bernstein analyst Geoffrey Porges said of the speedy approval. "We're seeing a fairly consistent pattern of drugs for high unmet needs diseases being approved on an expedited basis."

Bayer said Stivarga will be ready for distribution as soon as Friday and set a wholesale price for the drug of $9,350 for a 28-day cycle of treatment.

In pivotal clinical trials, Stivarga - known chemically as regorafenib - helped severely ill patients live on average 1.4 months longer than those who received standard treatment without the Bayer drug. Stivarga plus standard care also delayed tumor growth for an average of two months compared with 1.7 months for standard care alone.

Bayer is currently awaiting a colon cancer approval decision for the drug in Europe and is also seeking U.S. approval of regorafenib as a treatment for gastrointestinal stromal tumors.

"Stivarga is the latest colorectal cancer treatment to demonstrate an ability to extend patients' lives and is the second drug approved for patients with colorectal cancer in the past two months," Richard Pazdur, director of the Office of Hematology and Oncology Products in FDA's Center for Drug Evaluation and Research, said in a statement.

In August, the FDA approved Zaltrap, an advanced colon cancer treatment from Sanofi SA and Regeneron Pharmaceuticals.

Onyx Pharmaceuticals Inc is entitled to a 20 percent royalty payment on global net sales of Stivarga under a settlement agreement with Bayer following a dispute over how closely related the medicine is to Nexavar, the kidney and liver cancer drug that the companies co-developed and share.

Bayer and Onyx will jointly promote Stivarga in the United States.

Porges, Bernstein's biotechnology analyst, called the Onyx royalty agreement, "a fairly attractive deal."

But he said initial sales expectations for Stivarga are relatively modest "because this is really a relatively small indication late in the disease."

However, Morningstar analyst Damien Conover forecast sales of the drug reaching $1 billion by 2020. Bayer declined to provide its own sales forecast for the medicine.

"Bayer has very high expectations for Stivarga," Shannon Campbell, head of oncology for Bayer Healthcare, said in a telephone interview. "We certainly expect that Stivarga is going to play an important role once (patients) have gone through some of the more standard infusion based chemotherapies that are out there."

Stivarga is a pill that works by blocking several enzymes that promote cancer growth.

The drug was reviewed under the FDA's priority review program that provides an expedited six-month review for drugs that offer major advances in treatment or that provide treatment when no adequate therapy exists. A decision had been expected by October 27.

The Stivarga label will include a warning that severe and fatal liver toxicity occurred in some patients treated with the medicine during clinical studies, the FDA said.

In addition to Zaltrap, Stivarga will likely also compete with Roche Holding AG's multibillion-dollar cancer drug Avastin and Erbitux from Bristol-Myers Squibb Co and Eli Lilly and Co.

An estimated 143,460 Americans will be diagnosed with colorectal cancer, and 51,690 will die from the disease in 2012, according to the National Institutes of Health.

Bayer shares rose 1.3 percent to 68.40 euros in Germany.

Onyx shares were up 2.8 percent at $84.85 on Nasdaq.

EMBL Chemical Biology: George Whitesides on Ligand Binding

Email This Entry

Posted by Derek

Now the conference day is winding up with a big talk by George Whitesides. He's talking about his thoughts on enzyme function, with reference to his group's work using carbonic anhydrase as a model. He praises its stability ("a ceramic brick") and other characteristics, as you might expect from someone who's published an entire review on its use in biophysical studies.

So what makes compounds bind to enzyme sites? His take on the hydrophobic effect is that he thinks it's due as much (or more) to changes in networks of water molecules, rather than just the release of structured water at the protein-ligand contact. The latter is important, for sure, but not the whole story. "There is no one hydrophobic effect", he says, "there are many hydrophobic effects".

Another quote: "There ain't nothin' like water", and I definitely agree. We're used to water, since it's the most common chemical substance that we deal with in our lives, but water is weird.

And there's a lot we don't know about it still. For example, Whitesides has just pointed out that we have a reasonable understanding of surface tension in the bulk phase - but not at all for molecular-sized holes. This is crucial for understanding ligand behavior. His view of protein-ligand binding, he says, is very water-centric. . .

No More Man Cave! ? Seng. DIY Projects, Scrapbooking and

After a long wait, our family now has an actual HOUSE! Woot woot.

If you recall, our first home was a huge shop with living quarters (e.g. a man cave) on about 6 acres of land that we were going to build our real home on. We lived there for four years ? people often ask me how a city girl could live in a ?garage? for that long. I tell them, ?Love makes you do some crazy things.?

We then got pregnant with Ansley, realized that the property was too far from everything and everyone and the fact that there weren?t any kids around that the new baby could play with. We moved out into a townhouse at the same time we were putting the shop on the market.

It took FOREVER to sell because of the new housing market regulations (and the fact that the property wasn?t a ?typical? house that the average Joe could get a mortgage on). We moved into a two bedroom townhouse in March of 2011 and finally sold it in February of 2012 to a guy who purchased it with cash.

Two babies later, on Ryan?s 30th birthday, we closed on a real home and are moving in this weekend! We are super excited.

This house buying journey was definitely a crazy one. Now that we had the experience of one house purchase under our belt, we thought we knew exactly what each of us wanted in a home (although we were never really in agreeance with really anything, but that?s a different story). We had been searching since the beginning of 2011 ? so its been a very long journey. We butted heads quite a bit; I cried on several occasions and guilt tripped him into making an offer on a house that he absolutely hated that had a child predator in the neighborhood (long story); I?ve made some realtors mad because I didn?t know the etiquette for asking questions about a property; and lastly, I learned a lot about short sales ? more about this later.

People that know me shouldn?t be surprised that I have a notebook with pictures and listing prices of all the houses we have been in or just drove by. My tally on this house search is up to:

  • 16 that we?ve visited with our realtor
  • 125 that we?ve driven by
  • too many to count that we?ve looked at online.

Recall our top three things that each of us wanted here. Ryan sort of got one of his things ? ?land?. Its on an estate lot ? 0.75 acres. Not really the ?land? for hunting he was wanting ? but we agreed we couldn?t afford the crazy 300 acres he was wanting at this point in his life. I got my top three =) I mean, his needs were kind of ridiculous when it boiled down to our price point. (That?s my way of not making myself feel bad because he didn?t get his other two things.) We agreed to open a savings account for his ?land? fund.

Anyway, this house we stumbled upon online looking in a city that we weren?t even considering. Its 15 minutes outside the city, but is a little city in itself. It was in a city that I was adamant about not living in until my realtor told me that one of her other clients said it was a great school district. The house is 5 minutes away from the nearest grocery store, which is closer than the nearest grocery store from our apartment. Its a town of 8,000 people. When we saw the house and the neighborhood it was in, we fell in love. When we saw the listing price, we fell in love even more ? until we found out it was a short sale. You see ? short sales take FOREVER. Usually, if you buy a house and contract on it ? it takes about three weeks to close. Short sales can take at the shortest two months to two years to close.

Our house was a short sale ? what this means is that the sellers OWED more than they could sell the house for (e.g. the market took a dump and what they originally purchased it for is not even worth the value they still owe on the mortgage). It takes forever because even when we did submit the offer and the sellers agreed to the contract, its the bank who has to also agree or counter because they are the ones taking a hit.

Anyway, after three FULL LONG months of waiting ? we got the bank to accept our offer. And the icing on the cake was that it appraised for more than we purchased for (considering we sold our first home at a loss ? this was a big win for us).

For those considering a short sale, here?s our example timeline to give you an idea of our wait time. Thankfully, we were on a month to month contract with our townhouse rental. The day numbers are calendar days from when I started counting our short sale transaction timeline:

5/19 ? Viewed Property and submitted offer
5/22 ? Signed seller?s disclosure
5/24 ? Day 1: Seller accepted the offer
6/06 ? Day 14: BPO was ordered
6/08 ? Day 16: BPO was completed
7/08 ? Day 45: Motion filed by seller?s lawyer for the court to approve the sale (due to seller being in chapter 13 bankruptcy)
7/23 ? Day 61: Bankruptcy court approved the sale, packet has now been sent to the negotiator
8/03 ? Day 72: Contract has been submitted for the investors (Fannie Mae) to review
8/13 ? Day 83: Contract has been approved by bank and the investors, given two weeks to close. Signed arms length affidavit.
8/20 ? Day 89: Called appraiser to let them know that appraisal must be done right away
8/21 ? Day 90: Appraiser goes out to view the property
8/27 ? Day 97: Appraisal sent to us, which starts 3 day waiting period before closing
8/30 ? Day 99: Closed

Our realtor even told us that she was surprised that the short sale went through. She also said, ?Someone must be looking down on you.? I asked her what she meant, and she said, ?You sold your property, had a healthy baby, closed on a short sale house on your maternity leave. What more can you ask for??

I agreed. And Ryan said, ?Well, I didn?t get my man cave.?

She said, ?You had four years of a man cave, and I don?t know how Seng put up with it.?

I agreed again.

If you or anyone you know needs a realtor, contact Cynda. Her team is truly awesome ? and so is her little Volkswagen Beetle. =)

No pictures for now ? but I?ll be posting more about the house and house projects soon enough. The first pictures will be of the before and afters of refinishing our hardwood floors. Stay tuned!
